The absence of a reliable standardized animal model, however, continues to impede advancement in this field. To address this issue, we propose an innovative strategy for remodeling the RISD microenvironment through a sequential process involving initial ROS neutralization, followed by suppression of further ROS formation and restoration of key cell proliferation pathways and functions. As proof of principle, we fabricated a composite microneedle (MN) patch using γ-polyglutamic acid as the supporting matrix and ruthenium (Ru) cluster-decorated magnesium silicate nanosheets (MSR NSs) as the bioactive enzyme-mimicking element. The incorporated Ru clusters display strong ROS-scavenging performance and activate the peroxisome proliferator-activated receptor signaling pathway, as demonstrated by transcriptomic sequencing. Concurrently, physiological degradation of the magnesium silicate component releases magnesium and silicate ions that promote cellular proliferation, migration, and new blood vessel formation. Using a radiation-induced skin defect animal model, we compared the reparative efficacy of the MSR@MN patch against both a γPGA-MSR ointment formulation and the commercial agent Orgotein. Results indicate that the MSR@MN patch successfully corrects the dysregulated microenvironment associated with excessive ROS, dampens inflammation, and facilitates mature vascularization along with effective tissue reconstruction.
